17-Beta Estradiol and Prednisolone as Potential Stimulators of Hair Regrowth in Chemotherapy-Induced Human Hair Follicle Damage via 'Dystrophic Catagen' Promotion

The study explored the potential of 17-beta estradiol (E2) and prednisolone to stimulate hair regrowth in human hair follicles (HFs) damaged by chemotherapy, specifically through the promotion of the 'dystrophic catagen' pathway. This pathway, while initially causing greater alopecia, was associated with faster recovery of HFs. Using an in vitro model, the researchers found that E2, when administered with a cyclophosphamide metabolite (4-HC), shifted human HFs into this pathway, enhancing pigmentary disturbances and inhibiting hair growth by reducing proliferation and increasing apoptosis of matrix keratinocytes. The effects were further enhanced by the addition of prednisolone. These findings suggested that E2 and glucocorticoids could potentially be used as a clinical tool to accelerate hair regrowth in chemotherapy-induced alopecia (CIA) in humans, similar to effects observed in murine models.
