    This study explored the role of melanocyte stem cells (McSCs) in melanoma formation using a c-Kit-CreER-driven mouse model. It demonstrated that oncogenic McSCs could indeed be a source of melanoma, expanding within their niche and forming invasive epidermal melanomas. The study found that normal niche signals, such as Wnt and Endothelin during hair growth phases, were co-opted to facilitate McSC malignant transformation. Additionally, the molecular profiling of McSC-derived melanoma in mice showed a strong resemblance to human melanoma in terms of heterogeneity and gene signatures. These findings provided experimental support for the human melanoma progression model and offered insights into the transformation and diversity of McSC-derived melanoma.
