Chronic Graft-Versus-Host Disease Exhibits Distinct Th1-Skewing and Barrier Abnormalities in the Skin

    This study investigates the molecular mechanisms of chronic graft-versus-host disease (GvHD) in the skin by analyzing the transcriptome of skin biopsies from 10 GvHD patients, 12 with atopic dermatitis (AD), and 10 healthy controls. The research identifies a strong Th1 polarization in GvHD, marked by genes like CXCL9/10/11 and IFN-γ, compared to controls and AD. In contrast, AD shows significant Th2-skewing with genes such as IL-4R and IL-10/13. Both GvHD and AD exhibit barrier abnormalities, but with distinct differences; GvHD shows enrichment of barrier markers not seen in AD. The findings highlight differential Th1/Th2/Th17 polarization and unique barrier signatures in these inflammatory skin diseases, offering insights into their molecular profiles.
