Using a RP-C18 column and a specific mobile phase, the method achieved CLO elution at 10.1 minutes with high resolution from skin matrix interferences. Validation followed International Conference on Harmonization guidelines, ensuring selectivity, linearity (0.5–15.0 μg/mL), robustness, precision, accuracy, and detection limits (0.02 and 0.07 μg/mL). The method was successfully applied in in vitro skin permeation and hair follicle deposition experiments, proving its effectiveness for CLO quantification.
