This study investigated the effects of acute UVB exposure on retinoid metabolism in the epidermis and its role in the progression of cutaneous squamous cell carcinoma (cSCC). Using SKH-1 mice, researchers found that UVB exposure increased the expression of retinoid synthesis, degradation, and binding proteins, suggesting enhanced retinoic acid synthesis in the epidermis. These proteins were found to concentrate in more differentiated epidermal layers. The study also demonstrated that retinoic acid synthesis is crucial for normal UVB repair, as shown by changes in proliferation and differentiation markers with disulfiram treatments. Additionally, the expression of proteins STRA6, DHRS9, and ALDH1A1 varied throughout cSCC development, indicating their potential role in retinoic acid synthesis changes. These findings provided insights into why some retinoid treatments were effective against cSCC and could aid in developing better prevention and treatment strategies.
