The study investigated the role of 5α-reductase (5αR) in the behavioral complications of acute sleep deprivation (SD) in rats, focusing on its impact on neuroactive steroid levels in the brain. It was found that SD increased 5αR expression and activity, particularly in the prefrontal cortex, leading to elevated levels of the neurosteroid allopregnanolone. The administration of the 5αR inhibitor finasteride reduced SD-induced behavioral issues such as PPI deficits, hyperactivity, and risk-taking behaviors, similar to the effects of antipsychotic and mood-stabilizing drugs. Additionally, while allopregnanolone worsened PPI deficits, progesterone improved them in SD-affected rats. These findings suggested that 5αR played a significant role in the psychosis- and mania-like symptoms associated with SD by altering neuroactive steroid levels in the cortex.
