Stable Transfection and Identification of a Hair Follicle-Specific Expression Vector of IGFBP-5 in Goat Fetal Fibroblasts

    The study focused on the construction of a hair follicle-specific expression vector for IGFBP-5 in Inner Mongolia Cashmere goat fetal fibroblasts. Researchers created a vector, pCDsRed2-K-IGFBP5, by cloning IGFBP-5 downstream of the KAP6-1 promoter and inserting it into a vector containing a red fluorescent protein. This vector was transfected into goat fetal fibroblast cells using Lipofectamine 2000, and stable cell clones expressing red fluorescence were selected with Geneticin. Polymerase chain reaction confirmed the stable integration of the transgene. The findings indicated that this method was effective for constructing hair follicle-specific expression vectors and obtaining stable transfection donor cells for nuclear transfer.
