Follicular Delivery of Spironolactone via Nanostructured Lipid Carriers for Management of Alopecia

    Shamma RN, Aburahma MH
TLDR Nanostructured lipid carriers can effectively deliver spironolactone to hair follicles for treating hair loss.
The study explored the use of nanostructured lipid carriers (NLCs) for delivering spironolactone (SL) to treat androgenic alopecia, aiming to enhance its efficiency and safety by targeting hair follicles and reducing systemic side effects. SL-loaded NLCs were prepared using an emulsion solvent diffusion and evaporation method, resulting in spherical formulations with sizes ranging from 215.6 to 834.3 nm and an entrapment efficiency greater than 74%. The SL was found in an amorphous form within the NLCs, and the drug release exhibited an initial burst followed by sustained release. Confocal laser scanning microscopy demonstrated the potential for localized delivery of SL into scalp hair follicles, indicating a promising approach for treating hair and scalp disorders.

Crushing spironolactone pills and combining them with minoxidil for topical application may be effective for hair loss, with the addition of finasteride suggested. Adding rosemary and peppermint oils can help mask the unpleasant smell.

Spironolactone can remain effective for a long time in treating androgenetic alopecia, but it may not completely stop hair loss. Hair loss treatments like spironolactone, finasteride, and dutasteride slow down hair loss rather than cure it, and their effectiveness can vary based on individual response and the aggressiveness of the condition.

Spironolactone is more potent and lowers testosterone and DHT, while dutasteride only impacts DHT. For female hair loss, checking hormones and considering treatments like oral minoxidil, spironolactone, or topical minoxidil is recommended.
