Spherical Nucleic Acids as Emerging Topical Therapeutics: A Focus on Psoriasis

    The document from 2020 explored the use of liposomal spherical nucleic acids (L-SNAs) as a new topical treatment for psoriasis, emphasizing their ability to deliver antisense oligonucleotides to target specific inflammatory pathways. Preclinical models demonstrated that L-SNAs could penetrate the skin and effectively reduce IL-17 receptor expression, with a 57% reduction in IL-17RA mRNA in mouse skin and up to a 72% reduction in treated human 3D skin cultures. Despite the challenges posed by the psoriatic skin barrier, these findings provide a proof of concept for L-SNAs as a potential treatment for psoriasis and other skin diseases, pending further clinical trials to determine optimal dosing and application frequency.
