The study investigated the effects of diazepam (DZP) on anxiety and memory in male and female rats, focusing on the influence of the estrous cycle and progesterone. It found that the anxiolytic and amnesic effects of DZP were dissociated in female rats, unlike in males. Additionally, these effects were dependent on the estrous cycle phase, with females in proestrus not exhibiting these effects at a 2 mg/kg dose. The co-administration of a progesterone receptor antagonist restored these effects, highlighting the role of hormonal state in DZP's therapeutic and adverse effects. This is significant given the higher prevalence of anxiety disorders in women. The study involved 182 Wistar rats.
