The study presented a novel method for the topical delivery of all-trans retinoic acid (ATRA) by covalently bonding it to polyvinyl alcohol (PVA), creating a water-soluble amphiphilic nanomaterial. This PVA-bound ATRA acted as a pro-drug, allowing for sustained and controlled release of active ATRA, reducing adverse side effects such as skin irritation and hair loss. In vitro tests showed that active ATRA was released for up to 10 days, and in vivo tests demonstrated reduced inflammation and retention of the drug at the application site for up to six days. This approach significantly enhanced dermal accumulation of ATRA in explant pig skin.
