Pyruvate Kinase M2 Accelerates Cutaneous Wound Healing via Glycolysis and Wnt/β-Catenin Signaling

This study investigates the role of Pyruvate kinase M2 (PKM2) in cutaneous wound healing, highlighting its involvement in glycolysis and Wnt/β-catenin signaling. The research found that PKM2 expression and pyruvate kinase activity increased during wound healing in mice, facilitated by Wnt/β-catenin signaling. The use of TEPP-46, an allosteric activator of PKM2, improved human keratinocyte migration and wound healing. Additionally, co-treatment with TEPP-46 and KY19382, a Wnt/β-catenin signaling activator, significantly accelerated wound healing, as evidenced by increased expression of PCNA, keratin 14, and α-SMA, and promoted angiogenesis in wound beds. The findings suggest that activating glycolysis and Wnt/β-catenin signaling could be a promising therapeutic strategy for enhancing wound healing.
