Nano-PROTACs: State of the Art and Perspectives

    PROTACs are a promising new technique in drug development, offering high specificity and flexibility to target and degrade disease-relevant proteins. However, they face challenges such as low solubility, poor permeability, off-target effects, and metabolic instability. The integration of nanotechnology with PROTACs aims to improve their in vivo performance by addressing these issues. This review discusses advancements in PROTAC prodrug nanoparticle design, delivery systems, and loading approaches, as well as the split-and-mix approach for drug optimization. It highlights the potential of carrier-free nano-PROTACs and their combinational therapeutic effects, emphasizing their advantages and translational potential in achieving selectivity and controlled drug release.
