TLDR Targeting macrophages may improve wound healing.
The document reviewed the role of macrophages in skin wound healing, emphasizing their critical functions in various healing phases, including inflammation, proliferation, and remodeling. Macrophages were shown to regulate inflammation, promote tissue repair, and influence fibrosis through phenotypic switching. Dysregulation of macrophage activity, such as in diabetic ulcers, led to impaired healing due to persistent inflammation. The study highlighted the therapeutic potential of targeting macrophage activity and signaling pathways, such as NF-κB and interleukin signaling, to enhance wound healing, particularly in chronic wounds and diabetic conditions. Additionally, a phase III clinical trial with the macrophage-regulating drug ON101 showed promising results in improving healing in diabetic foot ulcers. Overall, the findings suggested that modulating macrophage responses could offer new therapeutic strategies for wound management.
