The study demonstrated that the combination of letrozole (LTZ) and quercetin (Qu) loaded into spanlastics significantly enhanced cytotoxic effects against MCF-7 breast cancer cells. Spanlastics, as surfactant-based elastic nanovesicles, provided high encapsulation efficiencies and sustained drug release, leading to superior in vitro cytotoxicity, cell cycle arrest, and increased reactive oxygen species (ROS) production compared to individual treatments and free drugs. The combination therapy showed synergistic effects, reducing cell viability more effectively and increasing apoptosis. This approach offered potential for improved breast cancer management through transdermal delivery, achieving high local drug concentrations with reduced systemic side effects.
