TLDR IMSG nanoparticles improve vaccine delivery and immune response through hair follicles.
This study demonstrated the potential of using inverse micellar sugar glass (IMSG) nanoparticles for transfollicular (TF) vaccination as a needle-free method. The IMSG nanoparticles showed enhanced stability and encapsulation efficacy for the model antigen ovalbumin (OVA) compared to other nanoparticle systems. When administered via the TF route, these nanoparticles achieved better follicular uptake and elicited significant humoral and cellular immune responses, especially when co-encapsulated with the adjuvant c-di-AMP. This highlighted the importance of optimized nanocarriers and adjuvants for effective TF vaccination. The study underscored the potential of TF vaccination using nanoparticles to deliver antigens through intact skin, offering a promising alternative to traditional vaccination methods.
