The study explores the use of carboxymethyl chitosan-silk nanofiber (CCSF) composite hydrogels loaded with forskolin to enhance skin regeneration. The hydrogels are designed for sustained release of forskolin, a compound known for its wound healing properties. In vitro tests showed that the hydrogels are non-toxic and promote the proliferation of human umbilical vein endothelial cells and mouse dermis fibroblasts while reducing oxidative stress. The hydrogels also inhibited myofibroblast migration, suggesting reduced scarring. In vivo results demonstrated accelerated wound closure, improved neovascularization, and enhanced hair follicle regeneration. This innovative approach shows promise for improving wound healing and skin repair.
