Effects of Exosomes Derived from Dermal Papilla Cells on Hair Follicle Stem Cells and Hair Follicle Organoids

    The study investigated the effects of exosomes derived from human dermal papilla cells (DPCs) on hair follicle stem cells (HFSCs) and hair follicle organoids using in vitro models. Exosomes were collected from three different culture methods: 2D, spheroid (3D), and spheroid microwell array cultures in an oxygen-permeable substrate (3D-oxy). All exosome types moderately promoted HFSC proliferation, but those from the 3D-oxy culture most effectively upregulated the expression of CD200, CD34, and K15 in HFSCs. Additionally, these exosomes were most effective in maintaining stem cells and stimulating hair follicle sprouting in vitro. The findings suggested that exosomes from human DPCs could have significant potential for treating hair loss.
