An Investigation of Keratin 15 Function by Small Interfering RNA Technology

    Cheryl Lancaster
TLDR Keratin 15 affects cell behavior and characteristics in skin cells.
The study investigated the role of keratin 15 (K15) in maintaining the stem cell niche by using siRNA to knock down K15 in various cell lines. It was found that K15 knock-down did not affect cell adhesion but did influence cell spreading, morphology, migration, differentiation, and proliferation. Specifically, K15-deficient cells spread more quickly, were larger, less motile, showed increased differentiation as indicated by higher involucrin expression, and proliferated more than control cells. K15 was present in the basal layers of the epidermis and around hair follicles in rat, mouse, and human skin. The study concluded that K15 impacts the stem cell characteristics of keratinocytes, though further research was needed to understand the underlying mechanisms.

Minoxidil with alcohol and propylene glycol was more effective than non-alcohol versions, and RU58841 worked best in a non-water-based solution. Oral finasteride showed better results than topical, despite side effects; microneedling and tretinoin worsened hair loss, and PRP was only helpful when younger.

The user replaced finasteride with saw palmetto due to availability issues and experienced side effects, leading to adjustments in their regimen. They use saw palmetto, minoxidil, Nizoral, and a dermaroller, reporting reduced side effects and some benefits, with hair fall ranging from 5 to 20 hairs daily.
