The study investigated the role of the neurosteroid allopregnanolone (AP) in the antihyperalgesic effect of paroxetine in a rat model of neuropathic pain. Paroxetine was found to increase AP levels in the spine, which contributed to its pain-relieving effects. The administration of finasteride, an inhibitor of AP synthesis, reduced the antihyperalgesic effect of paroxetine, indicating that AP plays a crucial role in mediating this effect. Additionally, intrathecal injection of AP alone produced similar analgesic effects as paroxetine, suggesting that therapies targeting AP synthesis could be promising for treating neuropathic pain.
