Formulation and Development of Adapalene Topical Nanohydrogel Using Different Surfactants and Cosurfactants for Anti-Acne Activity: In Vitro and Ex Vivo Evaluation

    TLDR NH-Tween-80 is a promising stable gel for treating acne.
    The study developed and evaluated an adapalene nanohydrogel (ADP-NH) using different surfactants and cosurfactants for anti-acne activity. The formulation with Tween-80 (NH-Tween-80) showed superior in vitro and ex vivo permeability compared to those with sodium lauryl sulphate (NH-SLS), glycerol (NH-glycerol), and a control solution. NH-Tween-80 demonstrated significantly higher adapalene permeation through both Strat-M® membrane and Wistar rat skin, and its ex vivo permeability was 3.38 times greater than a conventional market gel. Stability tests over 6 months showed NH-Tween-80 maintained 99.25% drug content without physical changes, unlike NH-SLS and NH-glycerol, which showed color changes. The study concluded that NH-Tween-80 is a promising stable formulation for treating acne vulgaris.
