The study found that Phospholipase Cδ1 (PLCδ1) was essential for skin stem cell lineage commitment, as PLCδ1-deficient mice experienced progressive hair loss, epidermal hyperplasia, and abnormal hair and skin development. The absence of PLCδ1 led to impaired calcium mobilization and protein kinase C activation, resulting in disrupted keratinocyte proliferation and differentiation. This caused sebaceous gland hyperplasia, cyst formation, and spontaneous skin tumors. The findings highlighted PLCδ1's critical role in maintaining skin homeostasis and preventing tumor development, suggesting its potential importance in developing treatments for hair loss and skin conditions.
