TGF-β Signaling During the Early Phase of Regeneration Is Required for Wound-Induced Hair Follicle Neogenesis

    TLDR TGF-β signaling is essential for new hair growth after a wound.
    This study investigates the role of TGF-β signaling in wound-induced hair follicle neogenesis (HFN) in mice. It was found that TGF-β signaling in fibroblasts during the early phase of wound healing is crucial for HFN, as it promotes the deposition of collagen type I in the wound dermis. Conditional knockout of TGF-β receptor 2 (Tgfbr2) or collagen type I (Col1a1) in fibroblasts inhibited HFN. However, activating the Sonic hedgehog pathway, essential for hair follicle development, could counteract the impaired HFN in collagen type I-deficient wounds. These findings suggest that TGF-β signaling is necessary for creating an environment conducive to HFN by ensuring adequate collagen type I deposition.
