The study investigated the effects of testosterone on vacuolar (V)-ATPase expression and activity in the vas deferens of orchidectomized rats. Testosterone treatment increased fluid secretion and decreased pH in the vas deferens, with these effects being antagonized by bafilomycin, a V-ATPase inhibitor. High levels of V-ATPase A1 and B1/2 proteins were found in the vas deferens epithelium of testosterone-treated rats. The study concluded that V-ATPase plays a crucial role in the acidification of vas deferens fluid under the influence of testosterone, which is essential for sperm maintenance and male fertility.
