TLDR Combining tadalafil and finasteride improves their solubility and effectiveness.
The study presents a novel coamorphous system of tadalafil (TDF) and finasteride (FNS) in a 1:1 molar ratio, aimed at enhancing the pharmaceutical performance of the USFDA-approved combination for treating benign prostatic hyperplasia. This system addresses the low aqueous solubility of both drugs by using mechanochemical synthesis to create a single solid phase. Characterization through various analytical techniques confirmed the formation and stability of the coamorphous phase. In vitro and in vivo studies demonstrated a significant improvement in dissolution and bioavailability compared to the physical mixture. The coamorphous system shows potential as a superior therapeutic alternative due to its improved solubility, dissolution, and pharmacokinetic properties.

A 20-year-old started taking finasteride for hair loss at 18, saw improvement, but developed erectile dysfunction (ED) after taking accutane. Despite stopping both medications, ED persisted, and hair loss worsened. Advice given includes considering tadalafil for ED, resuming finasteride, using minoxidil, and addressing psychological factors through positive thinking and lifestyle changes.
