The study presents a novel drug–drug coamorphous system of tadalafil (TDF) and finasteride (FNS) in a 1:1 molar ratio, aimed at enhancing pharmaceutical performance for treating benign prostatic hyperplasia. This system addresses the low aqueous solubility of both drugs by using mechanochemical synthesis to create a coamorphous phase, characterized by various analytical techniques. The coamorphous system demonstrated significantly improved in vitro dissolution and in vivo bioavailability in SD rats compared to the physical mixture. Stability tests confirmed its potential for industrial application, suggesting it as a superior therapeutic alternative to the existing marketed combination.
