Simultaneous Determination of Minoxidil and Minoxidil Sulfate by High-Performance Liquid Chromatography with UV Detection and Its Applications

    The study developed a high-performance liquid chromatography with UV detection (HPLC-UV) method to simultaneously detect minoxidil and minoxidil sulfate, with detection limits of 25 pg inj⁻¹ for minoxidil and 100 pg inj⁻¹ for minoxidil sulfate. It was found that minoxidil sulfate is unstable at high temperatures and should be stored at a maximum of 4 °C for HPLC analysis. The research also showed that arylsulfatases from human hair follicle cells and snail sources could convert minoxidil sulfate back to minoxidil, but this conversion could be inhibited by ascorbate-2-sulfate. This suggests a potential way to enhance the stability and effectiveness of minoxidil sulfate for AGA treatment. The method can be used to investigate minoxidil metabolism and find compounds that might improve minoxidil's bioavailability. Four accuracy control experiments were conducted, and the stability of minoxidil sulfate was tested over 22 hours at various temperatures.
