Screening of Sheep Fibroblast Cell Line with the Spider Dragline Silk Protein Gene

    TLDR Sheep cells were successfully modified to include a spider silk protein gene.
    Researchers synthesized a tetra-polymerized spider dragline silk protein gene and constructed a eukaryotic expression vector by linking it to a hair follicle-specific promoter. Sheep fibroblasts were transfected with this plasmid using a cationic liposome method and selected with G418. The transgenic cell line was confirmed through PCR, showing successful integration into the sheep genome. Chromosome analysis indicated stability and no differences from normal cells. This transgenic sheep fibroblast cell line was intended for future transgenic sheep studies.
