Production of Artificial Synthetic Spidroin Gene 4S-Transgenic Cloned Sheep Embryos Using Somatic Cell Nuclear Transfer

This study explored the production of artificial synthetic spidroin gene transgenic sheep cloned embryos using somatic cell nuclear transfer (SCNT). Researchers cloned a high glycine/tyrosine protein gene (HGT) promoter from sheep and constructed a eukaryotic expression vector, pcDNA3.1-HGT-4S, by linking it with the synthesized gene 4S. Sheep fibroblasts were transfected with this vector, and a transgenic cell line was established. These cells were used as nuclear donors to create embryos via SCNT, which successfully developed in vitro. PCR confirmed the integration of the synthetic spidroin gene 4S into the embryo genome. The study concluded that this method successfully produced transgenic sheep embryos with a hair follicle-specific promoter, though further research was needed to express synthetic spidroin genes in sheep hair follicles.
