The study developed a systematic method for the sensitive and specific determination of hair lipids, including trace amounts of intrinsic endogenous cholesterol, ceramide, cholesterol sulfate, and 18-methyl eicosanoic acid, using a combination of TLC/FID, LC/MS, and GC/MS. This method allowed for the characterization of hair lipid composition from the root to the tip of chemically untreated hair fibers from two females, demonstrating specific changes in hair lipids due to individual differences. The method aimed to clarify the roles of these lipids in the cell membrane complex of hair fibers, overcoming limitations of previous methods that struggled with sensitivity and precision in analyzing trace lipids.
