TLDR Olanzapine reduces stress-related anxiety in rats when given acutely.
The study investigated the anxiolytic effects of olanzapine in rats, hypothesizing that its action might involve the GABAergic system through the neuroactive steroid allopregnanolone (ALLO). Using the elevated plus-maze test, researchers found that acute administration of olanzapine (0.5 mg/kg) reduced anxiety-like behavior in stressed rats, but this effect was not observed with chronic treatment. The anxiolytic effect was negated when olanzapine was co-administered with finasteride (FIN), a 5-alpha-reductase inhibitor, suggesting that olanzapine's anxiolytic properties might be linked to its influence on steroid function and the GABA system.
