TLDR 1 alpha,25-dihydroxy-vitamin D3 and calcipotriol speed up cell differentiation in hair follicles.
The study investigated the effects of antipsoriatic compounds, specifically 1 alpha,25-dihydroxy-vitamin D3 and its derivative calcipotriol, on outer root sheath (ORS) cells from human hair follicles. These cells typically express hyperproliferation-associated keratins similar to psoriatic skin. In monolayer cultures, both compounds at a concentration of 10(-6) M completely inhibited ORS cell proliferation, with reduced effects at 10(-8) M. In organotypic cultures, the vitamin D analogues led to a thinning of the living cell compartment and a thickening of the horny layer, along with reduced expression of differentiation markers. The study concluded that these compounds primarily accelerated differentiation within the suprabasal living cell compartment without affecting ORS cell proliferation.
