The study utilized advanced light microscopy techniques, including FLIM, Airyscan, and 3D-SIM, to investigate hair damage and the role of keratin associated proteins (KAPs) in hair. It was found that FLIM could effectively quantify oxidative damage in hair by detecting the conversion of tryptophan to kynurenine. The study also revealed that the uptake of dyes into hair is influenced by the size and lipophilicity of the molecules, with a cuticle-cortex boundary potentially limiting penetration. The research provided insights into the subcellular localization of KAPs, which could inform future cosmetic treatments and the development of keratin-based biomaterials. Additionally, the findings might be relevant to understanding diseases unrelated to hair follicles.
