The study compared Quantitative Mass Spectrometry Imaging (QMSI) and LC-MS/MS techniques for evaluating drug penetration in skin tissues. QMSI, applied to human and mice tissues after topical application, provided detailed molecular images showing drug localization and concentration in different skin structures, such as hair follicles and sebaceous glands. The results demonstrated a 20-25% similarity with LC-MS/MS, validating QMSI's accuracy. Additionally, QMSI identified potential dermis contamination during sample preparation. The study concluded that QMSI, combined with histological staining, is a valuable multimodal approach for preclinical and clinical drug penetration studies.
