A Partially Automated Method for DNA Extraction from Marmoset Hair Follicles to Avoid Blood Chimerism

The study introduces a partially automated, non-invasive method for extracting DNA from marmoset hair follicles to address the issue of blood chimerism, which is prevalent in these primates. Tested on over 300 animals, the method uses proteinase K cell lysis and Promega’s Maxwell RSC system, successfully generating high-quality whole genome sequencing libraries with an average chimerism rate of 2.3%. This rate is comparable to blood-derived libraries, making hair follicles a reliable alternative for DNA sequencing. The study also notes that hair DNA is largely free from chimerism and closely resembles fibroblast DNA, which is less affected by chimerism. Additionally, the method is less invasive, reduces lab time, and is practical for species where blood sampling is difficult. However, there was an average of 3.5% bacterial contamination in the sequencing libraries, indicating potential environmental or fecal transfer.
