TLDR Twist2 is essential for scarless skin healing and hair growth in mouse fetuses.
The study investigates the role of Twist2 in skin regeneration and hair follicle formation in mouse fetuses. Twist2 expression was found to be upregulated in the dermis of embryonic day 13 (E13) wound margins but downregulated in E15 and E17 wounds. Knockdown of Twist2 on E13 resulted in visible marks, inhibited regeneration, and defective follicle formation. Twist2-knockdown dermal fibroblasts lacked the ability to undifferentiate, and Twist2 hetero knockout mice formed visible scars even on E13. These findings suggest that Twist2 is crucial for scarless wound healing and hair follicle regeneration, potentially guiding therapeutic strategies to reduce scarring and promote hair growth.
