This study explores the use of SH-SY5Y cell lysate as a substitute for brain homogenate in diagnosing Satoyoshi syndrome, a rare autoimmune disorder characterized by muscle spasms, diarrhea, and alopecia. Western blot analyses were performed on sera from three patients with Satoyoshi syndrome, 30 blood donors, and six patients with other neurological conditions. The results showed a consistent three-band pattern in the 70–100 kDa range for Satoyoshi syndrome patients across all substrates, which was absent in control groups. The SH-SY5Y lysate provided clearer and more reproducible results than brain homogenate, suggesting it could be a more effective and cost-efficient substrate for diagnosing Satoyoshi syndrome, potentially leading to earlier diagnosis and treatment.
