Rizatriptan Benzoate-Loaded Dissolving Microneedle Patch for Management of Acute Migraine Therapy

    TLDR Rizatriptan benzoate-loaded dissolving microneedles are effective and convenient for treating acute migraines.
    This study developed dissolving microneedles (MNs) using polyvinyl alcohol and poly (1-vinylpyrrolidone-co-vinyl acetate) for transdermal delivery of rizatriptan benzoate (RB) to treat acute migraines. In-vitro permeation studies and Franz diffusion cells demonstrated that these MNs enhance transdermal flux by effectively penetrating the skin. In-vivo pharmacological studies showed that RB-loaded MNs significantly alleviated migraine symptoms by modulating levels of 5-hydroxytryptamine, calcitonin gene-related peptide, and substance P. The study concludes that RB-loaded dissolving MNs offer safety, convenience, and high efficacy, supporting their potential as a transdermal drug delivery system for acute migraine treatment.
