Nano-Spanlastics-Loaded Dissolving Microneedle Patches for Ketotifen Fumarate: Advanced Strategies for Allergic Conjunctivitis Treatment and Molecular Insights

    This study presents a novel approach for treating allergic conjunctivitis (AC) using ketotifen fumarate (KF) loaded into spanlastics nano-vesicular carriers and incorporated into PVA/PVP dissolving microneedle patches. The optimized formulation demonstrated a small particle size, high zeta-potential, and entrapment efficiency, leading to a controlled 93% cumulative release over 72 hours with a zero-order kinetic profile. Pharmacodynamic evaluations showed significant reductions in inflammatory markers IgE, TNF-α, and IL-6, while increasing TGF-β and IL-10 levels, indicating effective anti-inflammatory effects. Gene expression analysis further supported the therapeutic potential, suggesting that this targeted, sustained release system could offer prolonged ocular therapy for AC.
