Study of Gonadal Hormone Drugs in Blocking Filovirus Entry of Cells In Vitro

TLDR Some gonadal hormone drugs can block filovirus entry into cells.
This study screened 1,600 commercial drugs to identify inhibitors of filovirus entry using a ZEBOV-GP/HIV pseudovirus model. Twelve gonadal hormone drugs showed inhibitory activity at a concentration of 10 μmol x L(-1). Notably, toremifene citrate, tamoxifen citrate, and clomiphene citrate exhibited strong inhibitory activities with IC50 values below 1 μmol x L(-1). Seven drugs, including estradiol benzoate and raloxifene hydrochloride, demonstrated moderate activity with IC50 values between 1 and 10 μmol x L(-1). Two drugs, hexestrol and chlormadinone acetate, showed weak activity. Additionally, toremifene citrate, tamoxifen citrate, clomiphene citrate, raloxifene hydrochloride, and quinestrol were effective against both Sudan ebola virus and Marburg virus pseudovirus entries.
