Mechanism of Transdermal Permeation Promotion of Lipophilic Drugs by Ethosomes

The study investigated how ethosomes enhance the transdermal permeation of lipophilic drugs, using Rhodamine B as a model drug. Ethosomes, composed of 2.45% yolk phospholipids, 30% ethanol, and 67.55% distilled water, significantly improved drug penetration into deeper skin layers compared to hydroethanolic solutions and liposomes. This enhanced penetration was due to the synergistic effects of ethosome ingredients and their ability to penetrate through hair follicles and the stratum corneum. Ethosomes were found to be superior carriers for transdermal drug delivery, with a longer retention time in skin appendages and the dermis, making them suitable for targeting hair follicles and sebaceous glands. The study concluded that the effectiveness of ethosomes was due to a complex interaction of their components, not just their alcohol or phospholipid content.
