The study investigates the age-related downregulation of dihydrotestosterone (DHT)-inactivating enzymes, specifically the aldo–keto reductase family 1 member C (AKR1C) family, in human scalp sebaceous glands (SGs). It was found that AKR1C expression, particularly AKR1C4, is strong in male SGs but declines with age. The androgen receptor (AR) is predominantly expressed in SGs and its localization is unaffected by age, sex, or hair loss condition. Sulforaphane treatment was shown to upregulate AKR1C expression in sebaceous gland cells and dermal papilla cells. These findings suggest that changes in SG steroid metabolism with age may affect local androgen metabolism and hair follicle homeostasis.
