October 2017 in “Proceedings of the National Academy of Sciences” The study highlighted the critical role of the zinc transporter ZIP10 in epidermal development, particularly in the context of zinc deficiency, which first affects the skin's epidermis. ZIP10 was found to be highly expressed in the outer root shelf of hair follicles and essential for proper epidermal formation. The absence of ZIP10 led to epidermal hypoplasia by reducing the transcriptional activity of p63, a key factor in epidermal morphogenesis. Conversely, ZIP10-mediated zinc influx enhanced p63 transactivation, promoting epidermal development. This research established the physiological importance of the ZIP10–p63 axis in maintaining epithelial homeostasis.
