October 2025 in “Current Issues in Molecular Biology” This study investigates the effect of escin on enhancing microcirculation in cutaneous tissue through topical application. Escin was found to stimulate the Wnt/β-Catenin and JNK signaling pathways in human umbilical vein endothelial cells (HUVECs), leading to down-regulation of glycogen synthase kinase-3 beta (Gsk3β). This down-regulation promotes angiogenic behavior and enhances blood vessel networks, resulting in improved cutaneous blood flow. The findings suggest that escin has potential as a modulator of cutaneous microcirculation, supporting its role in maintaining skin health by promoting vascular remodeling and increasing perfusion.
