November 2025 in “Biomolecules” This study investigates the role of FGF22 secreted by dermal papilla cells (DPCs) in regulating hair follicle stem cell (HFSC) proliferation and differentiation. Using a DPC-HFSC co-culture system, it was found that FGF22 overexpression in DPCs significantly increased FGFR1/FGFR2 mRNA expression and HFSC viability while reducing apoptosis. Conversely, FGF22 knockout decreased FGFR1/FGFR2 expression, reduced HFSC proliferation, and increased apoptosis. FGF22 promotes HFSC proliferation and differentiation by activating Wnt/β-Catenin, Sonic Hedgehog, and Notch signaling pathways, while inhibiting BMP pathways. These findings provide insights into potential applications in regenerative medicine and hair loss treatment.
