Key studies reveal the importance of chromatin dynamics, exosome function, and cell cycle inhibitors like p16INK4a and p21 in maintaining stem cell homeostasis. Quiescent hair follicle stem cells (HF-SCs) have unique chromatin landscapes, and exosomes are crucial for maintaining quiescence by modulating protein synthesis. Exercise-induced stem cell activation is affected by genetic factors, such as p16INK4a deficiency. The editorial also examines the effects of mitochondrial metabolism and caloric restriction on aging oocytes and hematopoietic stem cells. Despite progress, more research is needed to fully understand the molecular mechanisms of stem cell quiescence and develop therapies for age-related diseases.
