TLDR TRPA1 is crucial for mechanical sensitivity in skin sensory neurons.
The study investigated the role of TRPA1 in mechanotransduction in cutaneous sensory neurons using TRPA1-deficient mice. It found that while TRPA1 was not essential for acute cold sensitivity, it played a significant role in mechanical sensitivity. TRPA1-deficient mice exhibited significantly reduced action potential firing rates in C-fiber nociceptors and Ad-fiber mechanonociceptors in response to mechanical stimuli. TRPA1 was expressed in various neurons and keratinocytes, suggesting it modulated mechanotransduction through a cell-autonomous mechanism and interactions with keratinocytes. The findings highlighted TRPA1's crucial role in the mechanical responsiveness of cutaneous sensory neurons.
