The study developed a nanocrystalline suspension (NS) of dutasteride (DTS) for prolonged intramuscular delivery, using a dual-centrifugation bead-milling technique with Tween 80 as a stabilizer. The DTS-NS achieved a particle size of 324 nm and demonstrated a sustained drug release profile in rats, maintaining therapeutic levels for over 56 days with a bioavailability of approximately 67%. Histopathological analysis showed a granulomatous inflammatory response at the injection site, which significantly decreased by day 14 and was minimal by day 28. These findings suggest that the DTS-NS system is promising for sustained drug delivery with manageable local inflammation, offering potential improvements in the delivery and efficacy of DTS in clinical settings.
