The study focused on understanding the molecular mechanisms involved in adipogenesis and cellular plasticity, particularly in the formation of beige adipocytes. Researchers used innovative human ASC culture models and in vivo mouse experiments to explore these processes. They found that the Endothelial Growth Medium 2 (EGM2) preserved the immaturity of ASCs and enhanced their differentiation into beige adipocytes due to its low serum content, which reduced TGFß1 levels. Additionally, the protein SOX2 was identified as a key factor in promoting the formation of beige adipocytes and activating brown adipocytes, with its expression positively correlated with these processes both in vitro and in vivo. This research improved the understanding of culture conditions on ASC biology and highlighted molecules involved in adipocyte plasticity.
