Development and Validation of an HPLC-PDA Method for Quantitation of Ten Marker Compounds from Eclipta Prostrata and Evaluation of Their Protein Tyrosine Phosphatase 1B, α-Glucosidase, and Acetylcholinesterase Inhibitory Activities

The study developed and validated an HPLC-PDA method to quantify ten marker compounds from Eclipta prostrata, a plant used in traditional medicine for various ailments including hair loss. The method successfully identified and quantified compounds such as chlorogenic acid, quercetin, and apigenin. Compounds 5 and 6 showed significant inhibitory effects on the protein-tyrosine phosphatase 1B enzyme, while compounds 6, 7, 8, and 10 exhibited potent α-glucosidase inhibitory effects with IC50 values ranging from 23.8 to 45.5 μM. All ten compounds demonstrated considerable acetylcholinesterase inhibitory effects with IC50 values between 30.1 and 75.2 μM.
